East Fife defenders Mark McCulloch and Stevie Campbell and midfielder Robert Sloan miss the trip to Somerset Park.
Former Hearts and Airdrie United player Ryan Wallace could make his second appearance as a trialist.
Ayr United manager Brian Reid: "Our home form has been good for the majority of the season and I want us to get back on track straight away following the Livingston result (the 3-0 defeat).
"This will be a tricky game as East Fife will be boosted by holding Brechin City to a draw on Saturday."
East Fife manager John Robertson: "If we can come out of two back-to-back games against teams looking for promotion without losing a goal I am sure everyone will be delighted.
"It would give us a real platform for the rest of the season but as you would expect I also want us to show more threat up front."
